• Gastrointestinal bleeding, ulceration or perforation, which can be fatal, may occur at any time; risk is higher with higher doses, ulcer history and in the elderly. • Acetylsalicylic acid (aspirin): Irreversibly inhibits cyclo-oxygenase enzymes by acetylation, reducing prostaglandin synthesis (analgesic, antipyretic and anti-inflammatory effects) and, in platelets, blocking thromboxane A2 synthesis for the lifespan of the platelet (7-10 days), which inhibits platelet aggregation.
