• Most frequent (usually in the first 8 weeks): nausea, headache, arthralgia, mild gastric distress, skin rash and pruritus. • Use in pregnancy only after strict individual benefit/risk assessment at the lowest effective dose without added thyroid hormone; congenital malformations (e.g. aplasia cutis, choanal atresia, exomphalos, oesophageal atresia) are suspected, particularly with first-trimester and high-dose exposure; should be used in pregnancy only when propylthiouracil is not suitable.
