• Neuroleptic malignant syndrome-like reactions on abrupt dose reduction or withdrawal; impulse control/compulsive behaviors; caution in liver disease, recurrent eczematoid rash, congestive heart failure, peripheral edema, orthostatic hypotension, seizures, and untreated angle-closure glaucoma; do not stop abruptly in Parkinson's disease. • Not adequately tested for embryofetal development; in non-GLP rat studies amantadine increased embryonic death and severe visceral and skeletal malformations; human data are scarce (isolated reports of cardiovascular malformations after first-trimester exposure).
