• Levodopa: Crosses the blood-brain barrier and is decarboxylated to dopamine in the CNS, replenishing nigrostriatal dopamine.

• Entacapone: Inhibits peripheral COMT, prolonging levodopa plasma half-life when given with levodopa/DDC inhibitor.
